说明: 质谱级PNGase F 糖苷酶特异性水解复杂的糖蛋白的寡糖和高甘露糖链接的天冬酰胺,酶切掉天冬氨酸脱氨基的天冬酰胺,保留低聚糖蛋白完好无损。但是不会去除低聚糖含有的α(1→3)核心岩藻糖。

溶液酶解SOP:

1. 在 50mM Ammonium Bicarbonate (pH 7.8)溶液中加入20ug糖蛋白,调整体积至 18µl.

2. 加 2µl of 复溶的PNGase F

3.  37°C 保温 2–18 小时

活性:   40000unit/min/mg;

单位定义:PNGase F糖苷酶每分钟酶切1.0 umole Ribonuclease B的量,反应条件:37℃,PH=7.5

Description: Glycosidase PNGase F, Mass Spec Grade, (Peptide N-Glycosidase F) cleaves asparagine-linked high mannose as well as hybrid and complex oligosaccharides from glycoproteins. It deaminates the asparagine to aspartic acid, but leaves the oligosaccharide intact . PNGase F will not remove oligosaccharides containing α(1→3)-linked core fucose.

 

Physical form: PNGase F is supplied as a liquid in 20mM Tris-HCl , 50mM NaCl and 5mM EDTA at a concentration of 10,000u/ml

Storage Conditions: Store at 4°C-10°C, not freezed over -20°C

Shelf life:12 months at 4℃

Stability: PNGase F is maximally active in the pH range of 8.6 .

scope:PNGase F can effectively remove all N-glycans from both conserved and non-conserved  glyco-sylation sites.

 

In-Solution Protein Digestion:

1. Add up to 20µg of glycoprotein in 50mM Ammonium Bicarbonate (pH 7.8) to a final volume of 18µl.

2. Add 2µl of recombinant PNGase F

3. Incubate at 37°C for 2–18 hours

 

Activity:  40000unit/min/mg;

Unit Definition:One unit will catalyze the release of N-linked oligosaccharides from one nanomole of denatured Ribonuclease B in 1 minute at pH 7.5 at 37 °C. One  unit of PNGase F activity is equal to 1 IUB milliunit.
